Across-run comparisons of potency can be more variable than within-run comparisons (see Fig. 4). However, high-throughput screening responses can be affected by both random and systematic error and run-to-run variability should be not ignored 27. Obtaining experimental replicates can increase the precision of the potency estimates 28 and the interpretation of POD WES may be improved by focusing on robust assays with good agreement between compound measurements 29 and using appropriate signal curation 6. If potencies are only desired from a pre-specified functional form (e.g., the Hill model), a two-step procedure can be used to (1) find response profiles that are active according to a robust analysis framework designed to detect the desired trend 30 and then (2) estimate potencies from the returned profiles.

The AquaCure requires PURE water to operate. Many people buy their distilled water at a local store and that’s OK, but carrying gallons of water gets tiring AND costs more money than making your OWN distilled water. It doesn't matter how much water you use, it matters that there is no more than about 80 grams of lye in the machine. We compute the derivatives of WES using finite difference calculus, a mathematical procedure based on a Taylor series procedure that provides difference formulas for a grid sampled at discrete data points 32. If there are no detectable responses in the profile, the potency is declared “ undefined”. However, if potency cannot be estimated within the observed response profile but a detectable response is found within data region, finite difference calculus is used to predict the assay response beyond the tested concentration range. This extrapolation continues until POD WES is quantitatively estimated or designated “ less than C 1” for profiles that have substantial activity at the lowest observed concentration but no quantitative estimate is obtainable (see Fig. 1). No data points located within the detection window are extrapolated outside of the noise region. The estimation of POD WES is described in greater detail in the Supplementary Information. George Wiseman, the inventor of Eagle Research and the AquaCure systems, began his own research in 1986. He’s appeared on YouTube videos and many podcasts, and I credit him with today’s third resurgence of the subject. There’s no USA law that states any machine needs to be safety certified to be sold, but many business insurance plans require ALL equipment used in a building be safety certified. The function of the Humidifier is to filter out any residue of lye (NaOH) that comes out of the machine with the gas. It normally does a really good job. If there is not enough liquid or if it isn’t changed often enough, this function becomes compromised. Apply a continuous bead to studding and apply board immediately into position. Do not apply more than one board’s width of the stud frame at one time. Provide temporary support until cured.
