About this deal

Several studies have reported an increase in serum UMFA levels after implementing FA fortification, with possible concerns about potential overdoses and adverse effects. 1, 20 Variability in the presence or persistence of UMFA in the population that it may be accumulated in the blood due to different conditions, such as uncontrolled FA intake or impairment of the FA reduction pathway. High doses of FA can lead to UMFA syndrome, which is suspected of causing immune dysfunction and other adverse pathological effects, such as cancer, especially colorectal and prostate. 61-63 However, the causal effect between UMFA and those pathologies has yet to be demonstrated. 64 A Tolerable Upper Intake Level (UL), which refers to the maximum daily amount that can be taken without adverse effects, for folate from supplements or fortified foods is as follows:

For prophylaxis in chronic haemolytic states or in renal dialysis: 5mg every 1-7 days depending on underlying disease. Armitage JM, Bowman L, Clarke RJ, Wallendszus K, Bulbulia R, Rahimi K, Haynes R, Parish S, Sleight P, Peto R, Collins R. Effects of homocysteine-lowering with folic acid plus vitamin B12 vs placebo on mortality and major morbidity in myocardial infarction survivors: a randomized trial. JAMA. 2010 Jun;303(24):2486-94.

Key facts

In addition to dietary intake, plasma levels of folates are also modulated by genetic predisposition. Polymorphisms in genes coding for enzymes of the folate cycle are common—many of them with over 25% minor allele frequency (MAF) in the general population. The most critical are polymorphisms of the MTHFR (5,10-methylenetetrahydrofolate reductase) gene. MTHFR catalyses the conversion of 5,10-methylene-THF to biologically active folate, 5-Me-THF in an irreversible reaction [ 11]. The most important genetic polymorphisms that decrease the activity of MTHFR are two common functional polymorphisms, 677C>T (rs1801133) and 1298A>C (rs1801131), which are associated with reduced plasma levels of biologically active folate [ 12, 13]. Other polymorphisms in genes involved in folate uptake and metabolism could also affect the folate-homocysteine pathways and alter folate and/or homocysteine levels, including MTHFD1 1958G>A (rs2236225) [ 14], SLC19A1 80A>G (rs1051266) [ 15], MTRR 66A>G (rs1801394) [ 9], and others. Several studies report that individuals carrying these SNPs had altered levels of folate and/or homocysteine, although the results vary in the literature [ 16, 17, 18].

Folic Acid Tablets contain lactose. Patients with rare hereditary problems of galactose intolerance, total lactase deficiency or glucose-galactose malabsorption should not take this medicine. Another larger case series included 33 couples with fertility problems that had lasted for at least four years, such as recurrent fetal loss, premature ovarian insufficiency, or abnormal sperm parameters. Two-thirds of them had failed ART attempts. 52 For all the couples, at least one of the partners was a carrier of one of the two main MTHFR isoforms (C677T or A1298C), and the women had been previously treated unsuccessfully with high doses of FA (5 mg/day). The usual dose for adults and children is 5mg once a week, on a different day of the week to your methotrexate. Your doctor or midwife may recommend taking folic acid throughout pregnancy, particularly if you are at risk of anaemia or are anaemic. For the determination of intracellular concentrations of Hcy and 5-Me-THF, 10 million cells were pelleted and washed with 1 × PBS (pH = 7.4), and 150 µL of MiliQ water-containing antioxidants (DTT, ascorbic acid, and citric acid) was added to the pellet of cells and further processes as required, or stored at −80 °C before analysis. Frozen cell lysates were then thawed, and the internal standards of Hcy (4050 ng/mL) and 5-Me-THF (3 ng/mL) were added to each sample. The mixture was vortexed and sonicated (15 s, 40 mA), and then 750 µL ice-cold acetonitrile was added to each sample, vigorously vortexed (30 s), and left to stand on ice for approximately 20 min to allow protein precipitation. After that, the samples were centrifuged at 10,000× g for 10 min at 4 °C. The supernatants were transferred into new Eppendorf tubes and dried under a gentle stream of nitrogen at 40 °C. The residue was dissolved in 100 µL aliquots of MeOH:water (1:10), containing 100 µg/mL ascorbic acid, 100 µg/L citric acid, and 15 mg/mL DTT. The reconstituted sample was transferred to an autosampler vial with insert and subjected to LC-MS/MS analysis. The Agilent 1290 Infinity liquid chromatography system (Agilent Technologies, Santa Clara, CA, USA) and 6460 Triple Quad Mass Spectrometer (Agilent Technologies, Santa Clara, CA, USA) was used to analyze the samples of cell lysates. Chromatography separation was achieved on a Kinetex C18 column (100 × 3.0 mm, 2.6 µm) with a total run time of 12.10 min using a linear gradient of 0.05% acetic acid in water [ 32].

Prolonged exposure to high Hcy, hyperhomocysteinemia (HCys), can damage the blood vessels, contributing to blood clots, and can lead to the onset of cardiovascular disease, including atherosclerosis, stroke, and inflammatory syndromes as well as neuronal pathologies. Because healthy blood vessels and folate levels are essential for fertility and pregnancy, high Hcy can make it difficult to conceive and maintain a pregnancy. 25 Kirkland Signature Super B-Complex delivers not only 400 mcg of folic acid—the Recommended Dietary Allowance (RDA) for adults—but has the other B vitamins including thiamine, riboflavin, niacin, vitamin B6, vitamin B12, biotin, and pantothenic acid. It also provides 333% Daily Value of vitamin C and low amounts of electrolytes sodium and potassium. Alcoholism. Alcohol interferes with the absorption of folate and speeds the rate that folate breaks down and is excreted from the body. People with alcoholism also tend to eat poor-quality diets low in folate-containing foods. Although it's uncommon, vitamin B12 or folate deficiency (with or without anaemia) can lead to complications, particularly if you have been deficient in vitamin B12 or folate for some time.Taking too much folate from supplements or fortified foods can hide a vitamin B12 deficiency. Being deficient in vitamin B12 can lead to permanent brain and nerve damage. Taking too much folic acid may also increase the risk of some cancers, although more research is needed. Observational studies have been conducted with Quatrefolic in couples with fertility problems and have shown promising results. 22, 57 In Servy et al’s case series, seven couples with more than five miscarriages and a history of inefficient supplementation with high doses of FA (5 mg/day) to prevent pregnancy loss, were supplemented for four months with Quatrefolic. 57 Among seven couples, both partners in five carried the main MTHFR isoform (C677T). After the supplementation with 400 μg/day of Quatrefolic, six couples achieved pregnancy. Three were spontaneous pregnancies and deliveries, two had pregnancies at 16 and 25 weeks, and one had just started a pregnancy. Nutrition C for FS and A. It is not uncommon for supplement companies to pay labs for certificates after conducting minimal to no testing.
